On December 7, 1976, in New Orleans, Louisiana, to parents Cynthia (née Ernst) and Lawrence Duplass, Duplass entered the world. He went to Jesuit High School before enrolling at the University of Texas at Austin and then City University of New York, City College.

Both Jeff, Who Lives at Home, and The Do-Deca-Pentathlon, their 2012 flicks, were directed by them. Mark co-wrote, co-produced, and co-starred with Patrick Brice as Josef in the 2014 horror thriller Creep.
